Daily natural supplement to lower blood PAI-1 (a clotting-related protein)

Effect of a Daily Nitric Oxide-Stimulating Supplement on Plasma PAI-1 Levels: a Six-Week, Open-Label Clinical Trial

Phase1; Phase2 Interventional University of California, Los Angeles · NCT07469475

What this trial studies

Healthy adults without a history of acute or chronic cardiovascular disease will take a daily oral supplement containing ginger extract, L-citrulline, Paullinia cupana, and muira puama for six weeks. Blood will be drawn before and after supplementation to measure a panel of markers linked to vascular aging and inflammation, including PAI-1, IL-8, PCSK9, FGF-23, CRP, and Klotho. The study aims to see whether the supplement changes these biomarker levels in ways that could reflect improved endothelial function or reduced vascular inflammation. Participants who are pregnant, current smokers, using hormonal birth control, or allergic to the ingredients are excluded.

Who should consider this trial

Good fit: Adults aged 18 and older who do not have acute or chronic cardiovascular disease, are not current smokers, are not pregnant or using hormonal birth control, and are not allergic to the listed ingredients are ideal candidates.

Not a fit: People with prior serious cardiovascular events, active cancer (other than skin cancer), current smokers, pregnant individuals, those using hormonal birth control, or those allergic to the ingredients are excluded and unlikely to receive benefit from this study.

Why it matters

Potential benefit: If successful, the supplement could lower levels of clotting- and inflammation-related proteins and potentially support healthier blood vessel function as people age.

How similar studies have performed: Some individual components such as L-citrulline and ginger have shown modest improvements in endothelial function or inflammation in small studies, but this exact combination and formulation has not been widely tested in rigorous trials.
