What this trial studies

AMAZE 12 is a phase 3, randomized study comparing weekly subcutaneous NNC0487-0111 injections to a matching placebo in adults with excess body weight. Participants are assigned by chance to one of two arms and receive injections once a week with regular clinic visits for monitoring. Key exclusions include diagnosed diabetes, an HbA1c ≥6.5% at screening, and prior use of GLP-1–based treatments, dual GLP-1/GIP agents, or amylin analogues. Who should consider this trial

Good fit: Adults aged 18 or older with excess body weight who do not have diagnosed diabetes, have an HbA1c below 6.5%, and have not recently used GLP-1–based therapies or amylin analogues are the intended candidates.

Not a fit: People with type 1 or type 2 diabetes, an HbA1c ≥6.5%, or prior treatment with GLP-1/GIP or amylin medications before screening are excluded and unlikely to benefit from participation.

Eligibility criteria

Inclusion Criteria:

* Male or female (sex at birth).
* Age 18 years or above at the time of signing the informed consent.

Exclusion Criteria:

* Glycated haemoglobin (HbA1c) greater than or equal to ≥ 6.5% \[48 millimoles per mole (mmol/mol)\] as measured by the central laboratory at screening.
* History of type 1 or type 2 diabetes mellitus as declared by the participant or reported in the medical records.
* Treatment with glucagon-like-peptide-1 (GLP-1) receptor agonists (RA), dual GLP 1/gastric inhibitory peptide (GIP) RAs (or any other GLP-1 based treatment), or amylin analogues before screening.
