Comparing dry versus humidified incubators for sibling oocyte blastocyst development

Impact of Dry vs Humidified Culture Conditions on Blastocyst Development and Aneuploidy: A Time-lapse Sibling Oocyte Study

NA · ART Fertility Clinics LLC · NCT07270250

What this trial studies

This interventional sibling‑oocyte study splits oocytes from the same cycle between dry and humidified chambers of a GERI time‑lapse incubator under continuous culture conditions. Embryos are monitored by time‑lapse imaging and cultured to blastocyst stage, with outcomes recorded such as total blastulation, blastocyst quality, biopsy rate, fertilization rate, and aneuploidy after PGT‑A. The design compares matched sibling oocytes to control for patient‑level factors and isolate the effect of incubation humidity. Results aim to inform whether one chamber type yields better developmental outcomes while supporting uninterrupted lab workflows.

Who should consider this trial

Good fit: Ideal candidates are people aged 18–43 undergoing PGT‑A with ICSI who expect multiple mature oocytes (≥4), have BMI <35, and will use fresh or frozen ejaculated sperm.

Not a fit: People using PGT‑M or PGT‑SR, those using testicular sperm, undergoing conventional IVF insemination, or with a history of fertilization failure are excluded and unlikely to benefit from this protocol.

Why it matters

Potential benefit: If successful, the trial could identify an incubation approach that improves blastocyst development and helps labs choose methods that may raise IVF success while streamlining workflow and costs.

How similar studies have performed: Some small reports suggest humidified continuous‑culture conditions may improve blastocyst development in no‑refresh systems, but existing data are limited and inconsistent.
